What is S protein, why is it so important?
S protein is currently considered to be one of the most promising targets for the development of coronavirus vaccines and is being used in the development of vaccines against human coronavirus.


We could see from above diagram, the main structural proteins of 2019-nCoV are as follows:spike protein,envelope protein,membrane protein,nucleoprotein.

Among all of them, spike protein is the most important peripheral protein of 2019-nCoV, which has a high positive correlation with the virus’s infectivity. spike (S) glycoprotein alone can mediate the membrane fusion required for virus entry and cell fusion. It is also a major immunogen and a target for entry inhibitors. It consists of 2 subunits: S1 and S2. S1 mainly contains a receptor binding domain (RBD), which is responsible for recognizing the cell surface receptor. S2 contains basic elements needed for the membrane fusion. The S protein plays a key role in the induction of neutralizing-antibody and T-cell responses, as well as protective immunity.

Human anti 2019-nCoV(S) IgM/IgG ELISA Kit

This kit was based on indirect enzyme-linked immune-sorbent assay technology. Recombinant 2019-nCoV spike protein (antigen) was pre-coated onto 96-well plates. The test samples were added to the wells, unbound conjugates were washed away with wash buffer. Then added HRP conjugated anti-human IgM/IgG, if there were any anti-nCoV-IgM/IgG in the samples, it would form a complex. TMB substrates were used to visualize HRP enzymatic reaction. It was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The optical density of developed color is read with a suitable photometer at 450nm with a selected reference wavelength within 650 nm.
