Isotype control uses immunoglobulin and its subtype to match the primary antibody in the same species, Ig subclass and volume, removing background signal caused by non-specific antibody binding.
How to Choose Isotype Control
1. Use unlabelled primary antibody and labelled secondary antibody: the species and Ig subclass of isotype control antibody should be the same as primary antibody. When using labelled primary antibody, isotype control should have the same marker as primary antibody. Use the same testing concentration of isotype control as well to get the correct result analysis.
2. If suitable isotype control is not found, selection priority should be different subchain → different subclass → same category. (following the same species, fluorescence labelling and Ig subclass)
E.g. For a Mouse IgG2а,κ antibody, if the same isotype control is not found in the list, then choose the same fluorescence-labelled Mouse IgG2а → Mouse IgG2b → Mouse IgG2.
Applications
1. Flow Cytometry
Staining Method: Sample Tube: Primary Antibody + Sample; Isotype Control Tube: Isotype Control + Sample; Experimenters should use isotype control to keep the accuracy of flow cytometry.

2. Immunohistochemistry and Immunofluorescence
When the staining signal of isotype control is weaker than the specific antibody, the antibody binds with the target protein in the sample.
3. Immunoprecipitation
The figure below shows control IgG. Isotype control sample can only detect the heavy chain band without target protein band.
4. ELISA
When ELISA assay detects a specific antibody, isotype control can be used to eliminate non-specific binding.
