Background

Dextran sulfates (DSS) are anionic polymers produced by the reaction of dextran with chlorosulfuric acid, with a sulfur content of approximately 17% (equivalent to an average of 1.9 sulfate groups per glucose residue). In studies on inflammatory bowel disease (IBD) and colorectal cancer, DSS with molecular weights of 36,000–50,000 Da induces colitis in mice and rats by binding to chain fatty acids in the colon and triggering inflammation. DSS is a water-soluble polyanionic complex that forms a clear, colorless solution. It is commonly administered at 2%–5% directly in the drinking water of animals to establish disease models, or used at 3% in serum-free culture medium to stimulate cells to secrete inflammatory cytokines such as IL-6 and IL-10. However, when samples containing more than 0.3% DSS are used in ELISA assays, significant high background signals may occur, affecting data analysis. FineTest has developed a specialized blocking sample diluent (DSS Blocker Diluent) for high-concentration DSS samples, which effectively reduces background and restores assay accuracy.
