4',6-Diamidino-2-phenylindole dihydrochloride (DAPI) Staining Solution is a staining solution suitable for nuclear staining of all common cells and tissues. It can be used directly for nuclear staining of fixed cells or tissues. DAPI is a cell-membrane-permeable blue fluorescent dye that, upon binding to double-stranded DNA, exhibits fluorescence more than 20 times stronger than that of DAPI alone. Compared to EB (ethidium bromide), it offers significantly higher sensitivity for staining double-stranded DNA.

DAPI staining is commonly used in apoptosis detection, with observation under a fluorescence microscope or detection by flow cytometry after staining. In addition, DAPI is also widely used for routine nuclear staining and for double-stranded DNA staining under certain specific conditions.

The maximum excitation wavelength of DAPI is 340 nm, and its maximum emission wavelength is 488 nm. Upon binding to double-stranded DNA, the maximum excitation wavelength shifts to 364 nm and the maximum emission wavelength shifts to 454 nm.

This DAPI solution is prepared in water at a concentration of 1 mg/mL. For nuclear staining, the recommended working concentration of DAPI is 0.5–10 μg/mL.
