Immunostaining permeable solution(Triton X-100) is also called Immunostaining permeable buffer with Triton X-100, can be used for the permeability treatment of cell samples, frozen or paraffin sections during various in situ detections such as immunostaining, which helps expose the targets of action such as antigens and nucleic acids, making it easier for antibodies, probes or markers to enter the cells, thereby ensuring the detection effect of staining and other methods. This product is a ready-to-use working solution that can be used directly without dilution.

Permeable solution can usually achieve the permeability of cell membranes by using organic solvents such as methanol, acetone, etc., or by using detergents such as Triton X-100, Saponin, etc. Organic solvents such as methanol or acetone, on the one hand, can dissolve the cell membrane and nuclear membrane to fully expose the target proteins in the cytoplasm and nucleus, and on the other hand, can also denature the proteins within the cell and play a role in fixation. Organic reagents such as methanol or acetone are relatively easy to operate. One step of treatment can achieve both fixation and permeability simultaneously. However, the drawback is that membrane proteins can also be dissolved and some proteins denature, which is not conducive to subsequent detection. Therefore, organic solvents are used relatively less and are only used for some relatively rough detection requirements. Triton X-100 is a commonly used permeability reagent that can permeate the cell membrane and nuclear membrane. The principle of its action is also to dissolve the cell membrane non-specifically. Therefore, its drawback is that it is not conducive to the detection of membrane proteins. However, after cross-linking and fixation with paraformaldehyde, a considerable number of membrane proteins will be cross-linked and fixed, and thus will not be dissolved by Triton X-100. Therefore, it can still be detected later.

This Immunostaining permeable solution(Triton X-100) contains detergents such as Triton X-100 and is prepared in PBS. The immunostaining test results of the target proteins in the cytoplasm or nucleus of the samples treated with this immunostaining permeable solution (Triton X-100) show that the staining effect is basically the same as or slightly enhanced compared with the conventional permeable liquid.

Based on the calculation that each sample requires 0.1 or 1 milliliter of immunostaining permeable solution (Triton X-100), a 100 ml package of this product can permeate 1000 or 100 samples, and a 500mL package of this product can permeate 5000 or 500 samples.

Used for cell permeability in various routine immunofluorescence, immunohistochemistry, immunocytochemistry, flow cytometry detection, etc.

1.For sections, after fixation and washing are completed, 50-100μl of immunostaining permeable solution (Triton X-100) can be dropped on each sample. Permeation can also be achieved by complete immersion in the staining jar. For cell samples, after fixation and washing are completed, add 1 milliliter of immunostaining permeable solution (Triton X-100) to each well of a 6-well plate, and add it to other well plates in reference to this proportion.

For other samples, the amount of immunostaining permeable solution (Triton X-100) added should be sufficient to cover the samples.

2.Usually, the permeable process can be completed by incubating in the immunostaining permeable solution (Triton X-100) for 5 to 15 minutes at room temperature. For samples that are difficult to permeate or in cases where particularly thorough permeation is required, they can be incubated in the immunostaining permeable solution (Triton X-100) for 10-30 minutes at room temperature.

This product is only limited to scientific research personnel, can not be used for clinical diagnosis or treatment, food or medicine, must not be stored in the ordinary residential.
