The kit should be stored at -20℃ shading light for one year.
- Additional Materials Required
Cell or tissue samples, Precision Micropipettor, PBS buffer(PH7.3), Cover slip, slide
- Application
Used to slow down the fluorescence quenching of various common fluorescent dyes.
- Assay Procedure (For reference)
1.Preparation of anti-fluorescence quenching sealing solution:
After taking out the anti-fluorescence quenching reagent solvent, place it in a water bath at 30-40℃ for a short period of incubation. Then, add all the anti-fluorescence quenching reagent at one time to the anti-fluorescence quenching reagent solvent, fully dissolve and mix well before use. After preparation and after each use, it should be stored at -20℃ away from light. It can be used for at least three months.
2.Adherent cell sample:
A.After the dyeing is completed, suck up all the liquid.
B.Use a micropipettor to drop a drop of anti-fluorescence quenching sealing solution onto a slide, cover it with a cover slip containing the cells, and let the cells come into contact with the sealing solution. Try to avoid air bubbles.
C.Then the cell sample can be observed under a fluorescence microscope.
3.tissue section:
A: After the dyeing is completed, suck up all the liquid.
B.Use a micropipettor to drop a drop of anti-fluorescence quenching sealing solution on the tissue section, cover it with a cover slip, and let the tissue section come into contact with the sealing solution. Try to avoid air bubbles.
C.Then the tissue section can be observed under a fluorescence microscope.
4.Other samples:
Other samples should be operated with reference to cell sample or tissue section.
- Notes
1.Fluorescent substances are all prone to quenching. Samples after staining should be stored away from light. The use of anti-fluorescence quenching sealing solution can extend the fluorescence duration and slow down fluorescence quenching. However, the slide after sealing should still be stored at 4℃ away from light and taken pictures as soon as possible.
2.The anti-fluorescence quenching reagent should be added all at one time to the anti-fluorescence quenching reagent solvent, fully dissolved and mixed before use.
3.This product is only limited to scientific research personnel, can not be used for clinical diagnosis or treatment, food or medicine, must not be stored in the ordinary residential.
4.For your safety and health, please wear a lab coat and disposable gloves during the operation.
