Introductions

Accutase is a cell dissociation solution containing proteolytic and collagenolytic activities, serving as an ideal alternative to trypsin/EDTA solution for detaching cells from conventional tissue culture vessels and adherent culture systems. It is suitable for tissue dissociation and cell isolation, as the separated cells maintain intact surface antigens. Moreover, it is free of any animal- or bacterial-derived components, meeting the requirements of various downstream applications such as cell surface marker analysis, virus growth assays, flow cytometry, and bioreactor-related testing.

Compared to traditional trypsin digestion solutions, this product offers the following advantages:

① Suitable for digesting a wide range of primary cells and mammalian cell lines, including some insect cells;

② Gentle and effective digestion of stem cells (hESCs and mESCs), resulting in higher post-thaw recovery rates;

③ Rapid detachment of adherent cells within minutes, eliminating the need for washing or neutralization steps, thereby saving time during cell passaging;

④ Gentle on cells, leading to increased cell yield and viability, improved cell attachment efficiency, and enhanced cell morphology and growth characteristics.

Experimental Procedure

1.Thaw Accutase at 2–8°C or on ice, mix well by shaking, and aliquot for storage.

2.Rinse the cells once with calcium- and magnesium-free 1× Dulbecco's PBS.

3.Aseptically add 0.8 mL of Accutase to cover the adherent cells per 25 cm² surface area.

4.Return the culture to the 37°C incubator and digest for 2–10 minutes (the exact digestion time varies by cell type: monitor under the microscope until the cells shrink. Gently tap the culture vessel 3–5 times to detach cells completely).

5.After digestion, add the corresponding complete cell culture medium (volume determined by the size of the subsequent culture vessel) and mix gently by pipetting. Proceed to cell counting, passaging, or plating.

Notes

1.This product has been sterilized by filtration. Please maintain aseptic techniques during use.

2.Thaw the product at 2-8°C. Do not thaw at 37°C, and avoid prolonged exposure to room temperature or long-term storage at 2-8°C, as these conditions may affect enzyme activity. Mix well before use, and avoid repeated freeze-thaw cycles. For small-volume usage, aliquot and store frozen.

3.Digestion time can be adjusted based on the response of different tissues or cells to Accutase. It is recommended to monitor digestion progress under a microscope as a reference. No additional washing or enzyme inactivation steps are required.

4.This product is intended for research or further investigation use only, and is not for diagnostic or therapeutic purposes.
