- Background
This product can be used for the extraction of calprotectin from animal fecal samples. The main components are: 0.1M Tris, 0.015M NaCl, 1.0M Urea, 1.0mM CaCl2, 0.1M Citric acid, 5 mg/mL BSA, 0.25% Gentamycin sulfate, pH 8.0. Protein samples obtained with this extraction buffer were suitable for ELISA detection.
- Method of application
1.Try to collect dry stool, too thin stool will reduce the accuracy of the test. The weight collected should be greater than 100mg.
2.5 mL extraction buffer was added to 100 mg sample and vortexed for 10 min.
3.The samples were centrifuged at 5000×g for 10 min, then filtered through a 0.2 μm filter, and the supernatant was removed for detection. Or frozen in aliquots at -20℃ or -80℃ for later use.
- Notes
Calprotectin in stool remains stable for about 6 days, and it is recommended that unextracted stool samples be stored at 2-8 ° C for no more than 48 hours. It can be stored for a long time at -20℃. Samples were allowed to thaw slowly (overnight at 2-8 ° C) and equilibrate to room temperature before extraction.
