Abstract: IL-1F7 (IL-37, FIL-1 zeta, IL-1H4, IL-1HL, or IL-1RP1) is the anti-inflammatory member of interleukin-1 family. IL-1 family members are CNS mediators of innate immunity and inflammation. They’re pleiotropic cytokines with local and systemic effects.

1. Interleukin-1 Function in Inflammation
Most of IL-1 cytokines have proinflammatory activity (e.g. IL-1α, IL-1β, IL-18, IL-33, IL-36α, β, γ). Some members are anti-inflammatory. They play an important role in the biology of multiple inflammatory diseases.
2. Cytokines in Inflammatory Disease
IL-37 has the immunosuppressive effect and anti-inflammatory activity. The overexpression of IL-37 is related to various inflammations or autoimmune diseases. It's reported that IL-37 has a higher lever for patients suffering from rheumatoid arthritis, SLE, gouty arthritis, Graves disease and inflammatory bowel disease. Besides, IL-37 is also invovled in the mechanism of HIV infection and tuberculosis.
3. Anti Inflammatory Cytokines
IL-37 contains five isoforms (IL-37a-e). The study shows only IL-37b has the biological function and anti-inflammatory effect against various diseases. Usually, constitutive expression in healthy tissues is low. The expression in inflammatory counterpart is high, including synovium of patients suffering from active rheumatoid arthritis. Isoform a-c is expressed in testis, colon, placenta, lung and lymph nodes. Isoform d and e are just found in testis and bone marrow. Isoform a-c is found in brain, kidney and heart respectively.
Highly induced by LPS and TGFB1; Moderately induced by IFNG, IL18, IL1B, TNF and CpG. Due to the existence of IL-4 and CSF2, constitutive expression in bone marrow-derived macrophages is down-regulated. Induced by PMA in different cell lines. The stimulation of IL1B results in the colocalization with SMAD3, which occurs around perinuclear domain. Only the mature form cut by CASP1 is translocated into nucleus after the stimulation of LPS.
