Abstract: Nitric oxide(NO) is the autocrine and paracrine signalling molecule, which plays an important physiological role in promoting vascular dynamic balance. It can inhibit the contraction and growth of smooth muscle, prevent platelet aggregation, and avoid leukocyte-endothelial cell adhesion. Besides, it's also involved in immune defense system, neurotransmission, angiogenesis etc.

1. Nitric Oxide Cell Signaling
Nitric oxide(NO) is produced from the L-arginine oxidized by nitric oxide synthase (NOS). The half-life of NO is very short(about 5s). Thus, most researches on NO function are based on the regulation of NOS activity. Different types of NOS exist in various cells. Different mechanisms of signal transduction produce NO. Currently, there are three kinds of NOS, including endothelial tissue(eNOS or NOS3), nervous system(nNOS or NOS1), cytokine-induced type(iNOS or NOS2).
2. Biological Functions of Nitric Oxide
Downstream targets of NO include guanylate cyclase(cGMPase) and NF-κB. cGMPase can improve cGMP level. NF-κB functions as a key transcription factor in iNOS gene expression. The imbalance of NO level and signal in the body usually occurs in some diseases. Diabetic patients have the lowest NO level in the world. Atherosclerosis usually results in the damage of nitric oxide signaling pathway.
