How to prepare sputum sample while using FineTest® Elisa kit?

Sample Processing is to collect target protein. The processing shall be done as gently as possible as protein denatures or degrades easily. It is also very important to store processed sample, especially avoiding repeated free-thaw cycles. Processed sample can be sub-packed and sealed. Expiry Time: a week at 4 degree; a month at -20 degree; two months at -80 degree. Just put sample in room until its temperature reaches room temperature before using the sample. It is forbidden to heat the sample to melt the sample.

1. Select its thickness part and weigh it. Then add two times of sample weight of 0.1% DTT (dithiothreitol). Repeatedly blow it with a straw, and then vibrate it with swirler for 15 seconds. And later put it in 37 degree constant water bath and vibrate it for 5 minutes;
2. Put two times of sample weight of PBS in it and continue vibrating for 15-20 minutes. Then filter it with 150 mesh wire net, and centrifuge it at 1500rpm for 10 minutes and finally take supernatant and test it.
